CagriSema is a fixed-dose combination of Cagrilintide, a long-acting amylin analogue, and semaglutide, a GLP-1 receptor agonist. The combination is designed around the complementary actions of amylin and GLP-1 receptor pathways, which operate through distinct neural mechanisms within the hypothalamus and brainstem regions associated with appetite and satiety regulation.

GLP-1 receptor activity primarily influences appetite through hypothalamic and vagal signalling, while amylin receptor activation acts on the area postrema and nucleus tractus solitarius. This contributes to enhanced appetite suppression and delayed gastric emptying.
