AICAR

AICAR is considered a synthetic analog of adenosine monophosphate (AMP) precursors. While AMP is a nucleotide, AICAR functions as a cell-permeable nucleoside that, upon entering the cell, is phosphorylated into ZMP—a structural nucleotide analog of AMP. Because this metabolized form structurally resembles AMP, it directly activates AMP-activated protein kinase (AMPK), an enzyme central to regulating cellular energy balance by promoting energy availability while limiting excessive energy storage.

In experimental models, AICAR has been investigated for several potential research applications, including:

• Supporting insulin sensitivity by increasing glucose uptake by muscle tissue.
• Enhancing the breakdown and utilization of stored energy, particularly during ischemic conditions.
• Reducing intracellular lipid accumulation in tissues such as the liver.

AICAR may also influence fatty acid oxidation, mitochondrial biogenesis, and the preservation of slow-twitch, fatigue-resistant muscle fibres.
